This medication is a GABA analog and binds to the voltage-gated calcium channels in the central nervous system. This causes a reduced release of neurotransmitters like serotonin, dopamine, and substance P thus slowing down nerve signals.

Dosage & Administration of Pregaba ER 330mg Tablet

Adult Dosage

Neuropathic Pain & Postherpetic Neuralgia:

• Initial: 150 mg/day
• Increase to 300 mg/day after 3-7 days
• Max: 600 mg/day after a 7-day interval
• Administer in 2 or 3 divided doses

Diabetic Peripheral Neuropathic Pain:

• Initial: 50 mg PO q8hr
• Maintenance: Increase to 100 mg PO q8hr within 1 week, if needed
• Max: 300 mg/day

Adjunct in Partial Seizures:

• Initial: 150 mg/day
• Increase to 300 mg/day after 1 week
• Max: 600 mg/day
• Administer in 2 or 3 divided doses

Fibromyalgia:

• Initial: 150 mg/day
• Increase to 300 mg/day after 1 week
• Max: 450 mg/day, if needed
• Administer in 2 or 3 divided doses

Renal Dosage

Renal Impairment:

• Hemodialysis: 25-100 mg immediately after each 4-hour hemodialysis session

Creatinine Clearance (CrCl):

• 30 to <60 ml/min:
  • 75 mg/day
  • Max: 300 mg/day
  • Administer in 2 or 3 divided doses
• 15 to <30 ml/min:
  • Initial: 25-50 mg/day
  • Max: 150 mg/day
  • Administer as a single dose or in 2 divided doses
• <15 ml/min:
  • Initial: 25 mg/day
  • Max: 75 mg/day
  • Administer as a single dose

Interaction

• Angiotensin-converting enzyme inhibitors
• Benzodiazepines or narcotic pain medicines
• Thiazolidinedione antidiabetic agents

Contraindications

Known hypersensitivity to Pregabalin or its components.

Pregnancy & Lactation

• No adequate studies on pregnant women; and the potential risk to the fetus.
• Small amounts detected in human milk; breastfeeding is not recommended due to potential tumorigenicity.

Precautions & Warnings

• Not recommended during pregnancy or for those allergic to its ingredients.
• Consult a doctor if you have conditions like congestive heart failure, irregular heartbeat, diabetes, bleeding issues, muscle problems, high or low blood pressure, kidney problems, or a history of angioedema.
